安部研究室では、両生類の精巣をモデルとして培養系を用いて精子形成の分子機構を研究しています。両生類の精巣は培養が簡単であることと精巣構造が単純であるという利点があります。これまでに、濾胞刺激ホルモン(follicle-stimulating hormone; FSH)のみの処理によって精原細胞から第一精母細胞へ分化する器官培養系を確立しました。精原細胞の増殖因子としてSCFおよびIGF-Iが重要な働きをすることを明らかにしてきました。さらに最近、FSHとプロラクチンの濃度比が精原細胞の減数分裂開始か細胞死かを決定することを明らかにしました(減数分裂開始のチェックポイント機構)。
 また、温度や性ホルモン処理により容易に性転換を誘導することができるヒラメを用いて、性分化の分子機構を調べています。

 
 
 
 
In Abe's lab, we have been investigating the molecular mechanism of spermatogenesis using culture of amphibian testis as a model system. Amphibian testis has advantages that culture is easy and structure of the testis is simple compared with mammalianユs one. We have established organ culture in which addition of follicle-stimulating hormone (FSH) alone induces differentiation of spermatogonia to primary spermatocytes. Stem cell factor (SCF) and insulin-like growth factor (IGF)-I are shown to play important roles as growth factors in proliferation of spermatogonia. Furthermore, we recently clarified that concentration ratio of FSH/prolactin determines whether spermatogonia enter meiosis or go to die (checkpoint mechanism for entrance into meiosis).
In addition, we have been investigating the molecular mechanism of sex differentiation and sex reversal, using Japanese flounder of which genetic females can be sex-reversed to phenotypic males by rearing the larvae at high or low water temperature or by treatment with sex steroid hormones during the sex differentiation period.
