研究内容

1. 魚類(ヒラメ・メダカ)の性決定・性分化機構

 ヒラメとメダカの性は、通常は遺伝的に決定され、XX個体は雌へ、XY個体は雄へと分化します。しかしながら、性分化時期に性ホルモン処理を行うことで、容易に性転換を誘導することができます。特にヒラメは、性分化時期の高水温処理によりXX個体は雄へと性転換するなど、外的要因に対して大変感受性が高い実験動物です(原著論文 18)。そこで我々は、ヒラメを用いて性分化における性ホルモンの役割を調べました。ヒラメからアロマターゼ(アンドロゲンをエストロゲンに変換する酵素)、エストロゲンレセプター(ER)αとERβ、11β-水酸化酵素(魚類特異的アンドロゲンである11-ケトテストステロンの合成に関与する酵素)の遺伝子を単離し、温度コントロールにより誘導した雌雄の生殖腺におけるこれら遺伝子の発現を調べました(原著論文 13, 21)。その結果、アロマターゼ遺伝子の発現は、性分化開始以降、雌では急激に増加しましたが、雄では減少しました。ERαとERβ遺伝子の発現は、両者とも性分化時期における雌雄差は認められませんでした。一方、11β-水酸化酵素遺伝子の発現は、雄においてのみ、性分化後に増加しました。さらに、性分化時期のXX個体にアロマターゼ阻害剤(ファドロゾール)または抗エストロゲン剤(タモキシフェン)を投与することで雄へと性転換したことから、雌への性分化にはエストロゲンが不可欠で、雄への性分化にはエストロゲン合成の抑制が重要であると考えられました(原著論文 1, 19)。                    

 最近、我々はミュラー管抑制物質(MIS)の魚類ホモログの単離に成功しました(原著論文 6)。MISは、トランスフォーミング増殖因子β(TGF-β)スーパーファミリーに属する糖タンパク質であり、哺乳類、鳥類、爬虫類において、性分化時期に雄特異的な発現を示します。XXヒラメを18℃飼育により雌へ、11-ケトテストステロン処理または27℃飼育により雄へと分化誘導し、性分化時期におけるこれら雌雄の生殖腺でのMIS mRNAの発現をin situ hybridization法により調べた結果、性的未分化時期においてすでに性差が認められ、雄の生殖腺では強く、雌の生殖腺では弱いことがわかりました。また、性分化時期におけるMIS mRNAの発現は、雌の生殖腺では検出されず、雄の生殖腺の支持細胞でのみ観察されました。これらのことから、性分化時期でのMIS mRNAの発現パターンは、哺乳類、鳥類、爬虫類と同様に、ヒラメにおいても保存されていると思われます。魚類の性分化におけるMISの生理的機能はまだ明らかではないため、現在、MISの役割について詳細に調べているところです。

 一方、メダカは、小型でライフサイクルが短く、性を外観で確認することができるため、性転換する突然変異体を作製するために大変優れた実験動物です。我々は、原因遺伝子の同定が容易なエンハンサートラップ法を用いて、性転換の突然変異体の作製を精力的に行っています。また、いくつかの性決定・性分化関連因子を単離し、遺伝子ノックダウン技術を用いてそれら因子の機能解析を行うことにより、多大な成果をあげています。

2. 魚類(ヒラメ・メダカ)を用いた内分泌撹乱物質のモニタリングシステムの開発

 近年、多くの内分泌撹乱物質が環境中に存在することが明らかになり、生物への影響が懸念されています。例えば、今もなお沿岸域に存在する有機スズは、ヒラメXX個体を雄へと性転換させることが明らかになっています(原著論文 9)。そこで我々は、これら物質が生物に与える影響の分子メカニズムを明らかにし、これら物質のモニタリングシステムの開発を行う目的で、まず、エストロゲン様作用物質の作用機構を調べるためのレポーターアッセイシステムを構築しました(原著論文 2)。エストロゲン応答遺伝子であるビテロゲニン遺伝子のプロモーター領域とルシフェラーゼ(Luc)遺伝子を連結したベクター及びヒラメERαまたはERβの強制発現ベクターを培養細胞に導入し、Luc活性を調べました。その結果、ERαとERβのどちらを導入しても、E2の濃度依存的にLuc活性は上昇しました。次に、このシステムを用いて、ヒラメの雌化を誘導する3種類のエストロゲン様作用物質(ノニルフェノール、ビスフェノールA、ゲニスタイン)の作用機構を調べました(原著論文 2)。その結果、ノニルフェノール及びビスフェノールAは、ERβよりもERαを導入した方が明らかにLuc活性を誘導しましたが、ゲニスタインは、ERαとERβのどちらを導入しても同様にLuc活性を誘導したことから、ノニルフェノール及びビスフェノールAは主にERαを介して、ゲニスタインはERαとERβ両方を介してエストロゲン様作用を行うと考えられました。このシステムは、エストロゲン様作用物質の高感度なin vitro モニタリングシステムとして広く利用できると思われます。今後は、生体内で評価できるモニタリング用トランスジェニックフィッシュの作製を行っていきたいと考えています。
